Tratta dai fumetti di Garth Ennis e Steve Dillon, lo show segue le vicende del predicatore Jesse Custer (Dominic Cooper), che viene posseduto da una creatura soprannaturale. Genesis, il nome della creatura, ha in sé sia pura malvagità che pura bontà e ha una forza tale da poter affrontare Dio stesso. Custer, grazie a Genesis, possiede un potere inimmaginabile. Jesse insieme a Tulip (Ruth Negga), la sua ex findanzata, e a Cassidy (Joseph Gilgun), un vampiro irlandese, intraprende un viaggio alla ricerca di Dio dopo che quest'ultimo ha abbandonato il Paradiso dopo la nascita di Genesis.
